We largely know how PLK1 is activated in G2, but the players that regulate PLK1 in mitosis haven’t been fully described—a big gap in understanding how cell division is controlled. We find that Chk2, a DNA damage response kinase, has a novel role to sustain mitotic PLK1 activity

Although Chk2 and the DNA damage response pathway canonically antagonize mitotic kinases like PLK1 at the G2/M checkpoint, our data shows that this relationship is inverted in mitosis, highlighting how well-characterized interphase signaling pathways can be rewired in mitosis.
