Our lab investigates the molecular mechanisms that underpin the antibody-based cellular immune response to viral infection.

Scripps Research, La Jolla, CA
Why do some antibodies get NK cells to kill HIV-infected cells and others don't? We think it's geometry: nanometer-scale spacing of Env and bound antibody. New 5-yr NIAID R01 to measure it with MINFLUX, then design antibodies to spec. Hiring. Details 🧵

We're hiring: • Postdoc or staff scientist to lead the MINFLUX/super-resolution imaging • Technician for cell culture, cell assays, microscopy support — ideal for a recent or soon-to-be grad Interested or have leads? dmurin@sdbri.org. Institute ads posting soon.

Funded by a five-year, $4.7M R01 from NIAID/NIH (R01AI198054). This is our lab's second grant ever and by far the largest — a real milestone for a young group. With co-PI @ Daniel Montiel Garcia and co-Is Lars Hangartner & Andrew Ward at Scripps Research.

Then the inverse of how antibody discovery usually works: feed those measurements into ML models that predict which configurations drive the strongest ADCC, and use them to design synthetic nanobodies computationally. A blueprint instead of a lucky find.

Those distances are far below the resolution of conventional microscopy. To see them we'll use MINFLUX nanoscopy, which localizes single molecules to within a few nanometers. Direct follow-up to our recent work showing a key NK cell antibody receptor sits in pairs.

1/ Why do some antibodies get NK cells to kill HIV-infected cells while others barely work? Nobody's been able to predict it. Our hypothesis: geometry. The nanometer-scale arrangement of HIV Env and the antibodies bound to it on an infected cell's surface.
