It was a surprise to us that the Cia2 subunit of the targeting complex makes key interactions, as the current working model of CTC function delegates Cia2 to a Fe-S binding module. This discovery has 3 major implications for understanding this complex system.

1) the Cia2a paralog of Cia2 found in animals is possibly not a specific assembly factor for IRP1 but likely binds to and facilitates maturation of additional clients

2) we determine that a disease causing mutation in Cia1 results in disruption of the Cia1-Cia2 complex, and as a result, client recruitment and maturation

3) we find some of the first, direct biochemical evidence for role of the CIOA3/Nar1 protein in cluster trafficking. I hope you enjoy the paper! we are very excited to have this out there, if only in preprint form for now
