I know someone asked for the raw assay values that went into content uniformity: 132.16% 133.22% 108.60% 132.23% 134.40% 134.58% 134.61% 138.63% 135.30% 132.33% Based on this data, the 108 looks like a special cause (although not that special since the vial used for the assay result was 107%)

When we start running USP <788> on peptide vials, I fully expect to find high levels of glass and rubber particulates from unwashed vials. Peptide prices will rise as people learn the risks and demand testing. Paying $10 more per vial for clean product? Welcome trade.

It won’t just be the testing cost; the cost to manufacture will go up as well. Every failing test result has a bad process behind it. I think the vendors investing in quality processes now will reap the benefit of being ready as quality requirements increase.

USP 905 Content Uniformity is an FDA-USP test used to control dose-to-dose variation in dosage form (e.g. tablets, vials). It requires 10 assay values, and two things must be true for it to pass: 1. All the values have to be between 90-110% of the claim 2. The vial to vial variation has to be low enough that you can statistically predict all the vials will be in that spec

Why are there vials at 130% and some at 107%? Few things could cause this: > this is a mixed lot, even though I specifically requested all vials from one batch > there were two different pieces of filling equipment used to fill this batch > there was one piece of filling equipment with 8 filling heads and one is set wrong > equipment is malfunctioning

Purity is a great litmus test for how much method development went into a lab’s methods The lab with the lowest purity result has the most accurate method for that peptide If you buy peptides and you aren’t demanding a COA from the right labs… good luck

This is because of the science behind HPLC separations It takes work to make the chromatogram on the left. Sometimes weeks, sometimes months. Tweaking gradient parameters, trying different columns, etc. Any doof with an HPLC can make the chromatogram on the right in hours. Then use that same doofy method on 200+ peptides and get 99.9%+ purity on them all.
