リソソームの機能低下は老化を能動的に駆動する原因の一つであり、有望な創薬標的になりうる リソソームは細胞内の不要物を分解する細胞小器官であるが、加齢とともに生合成の低下、酸性度の低下、分解活性の低下、膜の損傷といった機能低下が生じることが明らかになってきた。リソソーム機能低下と老化・疾患との因果関係の解明、そして安全にリソソーム機能を回復させる治療法の確立が、依然として課題として残されている。 本レビューはリソソームの生合成と品質管理の分子機構を整理した上で、その機能低下が慢性炎症や細胞老化、さらには神経変性疾患・代謝疾患・感染症への脆弱性の増加にどのようにつながるかを論じ、リソソームを標的とした治療戦略の可能性を検討している。 具体的な治療戦略としては、軽度のリソソームストレスがかえって適応的な機能亢進を誘導する「リソホルメシス」という概念、転写因子TFEB/TFE3によるリソソーム生合成プログラムの活性化、そしてESCRT複合体を介した膜損傷修復機構の強化などが論じられている。 老化の「隠れたハブ」としてのリソソームの重要性を改めて示した本レビューは、今後の創薬標的としての注目を高めるものである。 nature.com/articles/s41556-0…
