Snuck onto the cover of Nature. Not a bad place for a “superdark” protein to end up. Very grateful. Much deserved Kyutae Lee, Sam Taylor, PhD, Jennifer Arcuri, Ph.D., Shraddha ChandThakuri, Bruno Colon, Jacinda Pujols, and others.
Out online today in @Nature. The known protein universe holds hundreds of millions of members, yet most remain unexplored, a vast biological darkness that may hold important clues to human health and disease. We went looking in it.

7th MCP Retreat ✔️
“Colab in the Cays” | Florida Keys
53 talks. 15+ labs. Every participant presenting.
Two days of deep science, new connections, and serious inspiration.
Thank you for the delicious birthday cake, Shraddha (the DAPI staining on the cells was spot on!), and gifts from Jen, Jacinda, Sam, and Kyutae❤️ Who doesn't love purple owls?
@MayorDaniella, this resilient elderly woman struggling to make it down the stairs because the elevator (visible) and down escalator at Dadeland South are broken. @FixMetroMDT @iheartmiami305
@MayorDaniella: this morning at Dadeland South. I didn’t take a photo of the disabled man arguing with security because he couldn’t get up the stairs. I am sure there is cc video of that if you want to see it. @FixMetroMDT@iheartmiami305
Winner winner chicken dinner! Great job on the poster, Sam, about acidic TMEs and illuminating the unknown. Thanks, @SylvesterCancer, for recognizing the hard work of our talented CAB graduate students. It's always a great retreat.
What a wonderful scientific retreat for UM Pharmacology this week @bakerscayresort. Time is running out to join our faculty and enjoy the Keys and science for yourself! bit.ly/3GAxDD6
I am first gen college and PhD. Thought I could connect/advise/listen to those in my shoes. So, I searched on Twitter, and this is what was returned. These are all excellent accounts, but this is it, save for one not shown.
Cool paper, but something about the pH measurement seems off. Washing yeast in water and resuspending in a buffer lacking glucose leads to acidification to pH ~6.2. This acidification is a super robust result. Perhaps the methods description is inaccurate or incomplete?
Congrats to our SURF/CREATE 2022 student Gorav Surana. A bright future lies ahead! Also, thanks again @TheRaiLab for creating and expanding this fantastic program that brings a perennially diverse cohort of undergraduates to Miami. @SylvesterCancer@umiamimedicine
Big shout out to our inspirational medical student @AlexanderRWeber@umiamimedicine. Such a positive presence in the lab and such an incredible job on your 6-week U SOAR project on opioid receptors. Thank you Dr. Grace Zhai for creating this wonderful program.
To confirm that our payloads are delivered, we do a gDNA prep and check by PCR prior to sequencing. An empty landing pad produces a PCR product of 223 bp with our primer setup. So, pretty small.
Jimmy, we are fans of the work, but one sentence in: "However, some receptors, including
the mu opioid receptor, are non-functional, which may be due to the presence of the fungal
sterol ergosterol instead of cholesterol". This is misleading. As for Fig. 1f, yes, from 1 year ago:
Every year I order a sheet cake for 50 to celebrate the culmination of my 8-week #pythonprogramming and #DataScience intensive here @umiamimedicine. I requested "GO PYTHON!" to be written on the cake. This is what I discovered this morning when I opened the box. Um, NAILED IT!😂
Although sample size is limited (n=2), the @danIsomLab is beginning to discover emerging trends in cork popping immediately following thesis defenses. @NKapolka @GeoffreyTaghon
Congrats to our past SURF student Sofia Perez on her graduation from the @NotreDame. Sofia will join us as a medical student at @umiamimedicine this Fall!
My recent talk in the #idg e-Symposium Series has posted to YouTube. I briefly overview findings we have recently published (and are soon to be published) in @PNASNews and @jbiolchem: piped.video/watch?v=pZWuVe4U…
To the UM community (and perhaps beyond), I will be hosting what promises to be an incredible seminar this Thursday 11/19 at noon sharp. Please join us!
An interesting collaborative project where we contributed our extensive expertise on the effect of buried ionizable residues on a protein structure-function relationship.

Congrats @_sofia_perez_9 on your 1st place win for your excellent SURF project and seminar. Way to represent the Isom Lab! Bright futures await you and your SURF cohort!
This week's #sinformaticsFactoids sticks with the GPCR theme and was inspired by the very recent melanocortin-4 structure from the Cone and Stevens labs (science.sciencemag.org/conte…), which has a calcium ion as a cofactor for ligand binding. Ergo, some fun with common ions...
The most prevalent heteroatom in human GPCR structures is the lipid oleic acid (OLA). There are 824 OLA molecules in the current set of 415 GPCR structures. Below, OLA binding hot spots identified by our consensus network analysis (CNA) algorithm #sinformaticFactoids.
Needed to update my list of current GPCR structures, but found my usual sources to be out-of-date or inaccurate. This prompted me to dust off some of my own informatics code. The result, 415 structure for 69 unique GPCRs. So begins my new regular series: #sinformaticFactoids.
