Somatic mutations accumulate in “normal” non-cancerous tissues (blood, skin, liver). But why are mutant clones selected? Do they merely proliferate more, selfishly? Or can clones adapt to disease? Could they protect tissues? We shed light on this: authors.elsevier.com/a/1gua9… 1/10

What gives some colorectal cancer cells the ability to metastasize? In our new paper, we find that tumor-intrinsic ceramide synthesis is a metabolic switch for metastatic initiation. 🧵 Free access: science.org/eprint/UAMCP9BAE…

New from Heidelberg University 🚨 In healthy liver, CYP2E1 expression is predominantly localized to zone 3 hepatocytes. In chronic alcohol-related liver disease, CYP2E1 expression reflects pericentral hepatocyte integrity rather than ethanol responsiveness, emphasizing the need for cell-type validation in liver disease studies. Timely, @Thelancet on GRAIL today! "To date, there is no evidence that any multicancer detection testing prevents cancer, consistently detects it at an earlier stage, or reduces cancer-associated mortality. Meanwhile, false-positive rates have been predictably high. The costs associated with evaluating false-positive results are very real, as are the psychological burdens borne by patients who are incorrectly told they have cancer—or incorrectly reassured that they do not." thelancet.com/journals/lance…
In its pivotal clinical trial, the GRAIL test to detect early cancer failed its primary endpoint. It detected only 3 early cases per 1,000 people assessed. There were more false negative results (N=196) than true positives (N=133). Reviewed here erictopol.substack.com/p/the… But you wouldn't know any of that if you read today's WSJ editorial! gift link wsj.com/opinion/a-grail-canc…

Why is glutathione, rather than cysteine, the major cellular thiol? Our new work by @ToshiNakamura51 suggests unlike other amino acids, excess cysteine is intrinsically toxic to mammalian cells via mitochondrial iron accumulation. @RockefellerUniv nature.com/articles/s42255-0…

Every so often, a major event happens in your life to validate years of work. SRRK’s ISEMBYLD was FDA approved. Having participated in its development from basic science to approval, as a physician-scientist, this is one of my best days. Happy for ISEMBYLD to help SMA patients.

How did we come to understand cancer in the light of evolutionary theory? Nothing makes sense if not with population genetics! In our latest @NatureRevCancer review we unpack how population genetics theory applied to somatic cells allows us to measure and predict cancer evolution from molecular data. Computational modelling in oncology has gone a long way but we need to go much further. The frontier is combining evolutionary theory with machine learning. Link below👇
